چکیده

This study aims to explore the clinical value of urine TWEAK/Fn14 mRNA in lupus nephritis (LN) patients. Thirty one SLE (Systemic Lupus Erythematosus) patients with or without active LN and another 10 healthy controls were recruited. LN activity was assessed by SLE Disease Activity Index. Urinary expression of TWEAK, Fn14 and MCP-1 at the mRNA level was measured by RT-PCR. Urinary expression of TWEAK, Fn14 and MCP-1 mRNA was significantly increased in LN patients (P<0.05). TWEAK, Fn14, and MCP-1 mRNA expression was positively correlated with 24-h urinary proteins, SLEDAI and serum anti-double stranded DNA antibodies (r=0.632, P=0.0001; r=0.651, P=0.001; r=0.417, P=0.02). TWEAK mRNA was positively associated with Fn14 and MCP-1 mRNA (r=0.871, P<0.0001; r=0.561, P<0.0001). In summary, TWEAK, Fn14 and MCP-1 mRNA are potential biomarkers of LN activity.

Urinary TWEAK Level as a Marker of Lupus Nephritis Activity in 46 Cases

OBJECTIVE This study is designed to observe the urinary tumor necrosis factor-like weak inducer of apoptosis (TWEAK) levels in patients with lupus nephritis (LN) and to identify new biomarker of lupus nephritis activity. METHODS Study subjects were 46 cases of patients with LN (including 34 of active cases) who underwent routine renal biopsy. Activity and chronicity indexes of LN were assesse...

Regulation of Tissue Responses: The TWEAK/Fn14 Pathway and Other TNF/TNFR Superfamily Members That Activate Non-Canonical NFκB Signaling

The immune system mediates tissue responses under both physiological and pathological conditions. In addition to leukocyte subsets, non-hematopoietic tissue cell types actively contribute to shaping tissue responses, including the inflammatory, fibrogenic, and regenerative components. TWEAK and its receptor Fn14, members of the TNF/TNFR superfamily, have emerged as a prominent molecular axis re...

TWEAK/Fn14 and Non-Canonical NF-kappaB Signaling in Kidney Disease

The incidence of acute kidney injury (AKI) and chronic kidney disease (CKD) is increasing. However, there is no effective therapy for AKI and current approaches only slow down, but do not prevent progression of CKD. TWEAK is a TNF superfamily cytokine. A solid base of preclinical data suggests a role of therapies targeting the TWEAK or its receptor Fn14 in AKI and CKD. In particular TWEAK/Fn14 ...

TWEAK/Fn14 interactions are instrumental in the pathogenesis of nephritis in the chronic graft-versus-host model of systemic lupus erythematosus.

TNF-like weak inducer of apoptosis (TWEAK), a member of the TNF superfamily, is a prominent inducer of proinflammatory cytokines in vitro and in vivo. We previously found that kidney cells display the TWEAK receptor Fn14, and that TWEAK stimulation of mesangial cells and podocytes induces a potent proinflammatory response. Several of the cytokines up-regulated in the kidney in response to TWEAK...

Urinary TWEAK as a biomarker of lupus nephritis: a multicenter cohort study

INTRODUCTION TNF-like weak inducer of apoptosis (TWEAK) has been implicated as a mediator of chronic inflammatory processes via prolonged activation of the NF-kappaB pathway in several tissues, including the kidney. Evidence for the importance of TWEAK in the pathogenesis of lupus nephritis (LN) has been recently introduced. Thus, TWEAK levels may serve as an indication of LN presence and activ...
